Immune Journey to the West

Another Encounter at Hidden Fog Mountain - The Peril of Drug-Resistant Antigen Relapse

Bidding farewell to the bustling Chen Village, the road once again wound into a vast, rolling expanse of deep mountains and ancient forests. Towering peaks and dense woods blocked out the sun, and a thick, lingering mist shrouded the area year-round. This was the infamous Hidden Fog Mountain. A thin, grey-black haze permeated the trees, and gusts of cold wind howled through the dense canopy.

No sooner had the group stepped into the territory of Hidden Fog Mountain than Sun Wukong (Monkey King) abruptly stopped in his tracks. He sniffed the misty mountain air vigorously, his brows locking tightly together.

"Something isn't right. I am far too familiar with this demonic aura. It's the Leopard Demon that used to wreak havoc here on Hidden Fog Mountain!"

Zhu Bajie (Pigsy) shivered from head to toe. He quickly hugged his Nine-Tooth Iron Rake tightly to his chest, his large ears trembling in fear as he hurriedly took two steps back.

"Brother Monkey, don't scare me! Didn't we teach that Leopard Demon a severe lesson back then? He was defeated long ago! How could he appear here again? Did this guy come back from the dead?"

Sha Wujing (Sandy) stopped walking, pinching a falling dead leaf between his fingers. He shook his head with a grave expression.

"Second Brother, I'm afraid it's not a resurrection. During our great battle back then, we only wiped out the majority of its main forces. A very small number of antigens managed to hide and lay dormant. Under long-term screening from immune pressure, they underwent genetic mutation and evolved into drug-resistant, immune-evading antigens. Our old immune memory is no longer capable of recognizing and dealing with them."

Before he even finished speaking, a villager in tattered clothes came stumbling frantically down the mountain road. He was dripping with sweat, his face pale and grey. He gasped for air, his expression etched with extreme terror.

"Help! The monster of Hidden Fog Mountain has come out to harm people again! Many of our townsfolk have contracted a severe illness. Their high fevers won't break, they can't stop coughing, and the medicines prescribed by the doctors aren't working at all! Some people have even suffered damage to their internal organs!"

Tang Sanzang quickly stepped forward to support the villager, who was about to collapse. His tone was full of concern. "Do not panic, benefactor. Take your time and tell us. Does the demon causing this chaos look like a leopard?"

The villager nodded repeatedly, his chest heaving violently.

"It is exactly the Leopard Demon! After you elders drove it away last time, it vanished without a trace. Who would have thought it would make a comeback now, and it is even more ferocious than before. The village doctors have exhausted all their methods, but the patients' conditions grow heavier by the day. Many people's organs have been damaged."

Tang Sanzang extended his fingertips, glowing with a faint light, to examine the villager's physical condition. A moment later, his face turned incredibly heavy.

"This is a relapse of a drug-resistant antigen. Back then, the residual antigens of the Leopard Demon hid inside the caves of Hidden Fog Mountain to lie dormant. Enduring long-term immune pressure, they underwent genetic mutation, and their antigen epitopes have changed. The antibodies and old effector T cells we generated previously have lost their ability to recognize it. A standard, conventional immune response is utterly powerless against it."

The First Prince looked utterly confused, scratching his helmet. "Master, does 'drug-resistant' mean the Leopard Demon has trained to become invulnerable, so much so that even Elder Sun's Golden Cudgel can't hurt it?"

Sun Wukong swung his glittering Golden Cudgel and grinned. "Kid, you're somewhat close. Being drug-resistant is like the monster putting on a brand-new invisibility cloak. My old combat tactics can't hit it; it can dodge our original immune attacks. But it's not like I have no way to deal with it. As long as we find the weak point of its mutation, I can still beat it until its soul shatters!"

Tang Sanzang immediately reached out to stop Wukong, who was eager to charge up to the cave.

"Wukong, you absolutely must not attack rashly. After becoming drug-resistant, the pathogen's virulence is even stronger, and our old immune weapons are entirely ineffective. If we attack blindly, it will only stimulate it to release more drug-resistant antigens and expand the scope of the infection. We must construct a brand-new adaptive immune response, specifically designed to recognize its newly mutated antigen epitopes. Only then can we break its drug resistance and completely eradicate this menace."

Zhu Bajie strongly agreed, nodding heavily. "Master is absolutely right. I can already feel that the antigens released by this Leopard Demon are completely different from back then. It's very difficult for me to drive an immune response using the old cytokines. We must develop an entirely new battle plan."

Tang Sanzang quickly issued combat orders. "Wukong, lead the three princes to the outskirts of the demon's cave on Hidden Fog Mountain to monitor their movements. You are only to observe; you are not allowed to start a fight without permission. Wait for us to complete the initiation of the new immune response. Bajie, prepare to differentiate brand-new Th1 and Th2 subsets to provide support for this novel immune response. Wujing, make preparations to generate specific antibodies targeting the mutated antigen epitopes at a moment's notice. White Dragon Horse, cast a mucosal barrier around the entire perimeter of Hidden Fog Mountain to block the drug-resistant antigens from spreading outward and to protect the villagers below."

"We obey your command!" The group received their orders and sprang into action immediately.

Without a word, Sun Wukong led the three princes and leaped into the dense, fog-covered forests of Hidden Fog Mountain. His Fiery Golden Eyes pierced through the heavy mist, quickly locking onto the deep, dark cave where the Leopard Demon was entrenched. The entrance was crowded with a large number of lesser demons infected by the drug-resistant antigen. Each had a ferocious appearance and was surrounded by an eerie, drug-resistant demonic aura.

The Third Prince lowered his voice, leaning close to Wukong's side, and whispered, "Elder Sun, these lesser demons at the entrance are all cells infected by the drug-resistant antigen. Why don't we just strike now and clear them out?"

Wukong gently shook his head, his eyes fixed on the cave entrance. "Absolutely not. Our old killing methods will have very little effect on these drug-resistant lesser demons. Striking rashly will only alert the enemy. We will wait until Master has fully initiated the brand-new response at the bottom of the mountain, and then we will coordinate from the inside and outside to annihilate them all in one fell swoop."

Meanwhile, at the foot of the mountain, Tang Sanzang activated the novel immune response mechanism. He collected a small sample of the drug-resistant antigen and precisely identified the mutated antigen epitopes. He processed the mutated antigen peptides, bound them to MHC I and MHC II molecules respectively, and presented them to Wukong and Bajie, while simultaneously releasing novel co-stimulatory signals.

"Bajie, differentiate your brand-new Th1 and Th2 subsets! Activate the immune troops! Wujing, prepare to generate brand-new targeted antibodies!"

Zhu Bajie took a deep breath, marshaling all the strength in his body to differentiate the new Th1 and Th2 subsets. The new Th1 released IL-12 and IFN-alpha, shouting loudly toward Wukong, who was monitoring the enemy in the forest. His voice pierced through the thick mountain fog.

"Brother Monkey! Activate your new effector CD8+ T cells! Your TCR needs to undergo adaptive modification to recognize the mutated antigen sites and break through its drug-resistant disguise!"

Following closely, the new Th2 released IL-13 and IL-14, turning to shout at Sha Wujing: "Brother Sha! Produce specific antibodies tailored exclusively for the mutated epitopes! Neutralize the drug-resistant antigens!"

Sha Wujing focused his energy, rapidly proliferating and differentiating into brand-new plasma cells and novel memory B cells. Batches of specific antibodies, like precision-guided missiles, poured out in a steady stream.

"Bajie, the new antibodies are in position and ready for combat at any time!"

Inside the mountain forest, Wukong received the new cytokine signals. The TCR receptors of his CD8+ T cells underwent adaptive changes, finally allowing him to accurately pinpoint the Leopard Demon's mutated antigen sites.

"Little demons, your invisibility cloaks are now useless! Watch how Old Sun deals with you!"

Zhu Bajie took a deep breath, marshaling all the strength in his body to differentiate the new Th1 and Th2 subsets. Activate your new effector CD8+ T cells! Your TCR needs to undergo adaptive modification to recognize the mutated antigen sites and break through its drug-resistant disguise!" Following closely, the new Th2 released IL-13 and IL-14, turning to shout at Sha Wujing: "Brother Sha! Produce specific antibodies tailored exclusively for the mutated epitopes! Neutralize the drug-resistant antigens!" Sha Wujing focused his energy, rapidly proliferating and differentiating into brand-new plasma cells and novel memory B cells. Batches of specific antibodies, like precision-guided missiles, poured out in a steady stream. "Bajie, the new antibodies are in position and ready for combat at any time!" The TCR receptors of his CD8+ T cells underwent adaptive changes, finally allowing him to accurately pinpoint the Leopard Demon's mutated antigen sites. "Little demons, your invisibility cloaks are now useless! Watch how Old Sun deals with you!" With a mighty roar, Wukong led the three princes and charged fiercely toward the cave. Hearing the commotion outside, the Leopard Demon roared and rushed out of the cave. "Sun Wukong! You've come knocking on my door again! I have now gained the power of drug resistance. Your skills cannot harm a single hair on my body! Just wait to be infected by me!" The Leopard Demon frantically released a massive wave of immunosuppressive factors, attempting to stifle Wukong's attack. "In your dreams!" Wukong sneered. The golden light of his Golden Cudgel surged as he smashed it violently toward the Leopard Demon. "Did you think you could run rampant just by relying on a mutated disguise? My brand-new set of killing skills was made specifically to deal with drug-resistant demons like you!" The brand-new specific antibodies produced by Sha Wujing arrived at lightning speed, directly neutralizing the immunosuppressive toxins released by the Leopard Demon, rendering them instantly ineffective. From the bottom of the mountain, Zhu Bajie continued to direct the battle. The new Th1 further activated macrophages, enhancing their phagocytic abilities to engulf the free-floating drug-resistant antigens everywhere. "Brother Monkey, step up the attack and eradicate it completely! We absolutely cannot leave any remnants behind again!" Wukong leaped high into the air, focusing all his power into the Golden Cudgel, and delivered a devastating blow squarely onto the critical mutated antigen epitope of the Leopard Demon. The Leopard Demon let out a heart-wrenching, agonizing shriek as its massive body shattered and disintegrated. The source of the drug-resistant antigen was completely destroyed. Without pausing, the group swept through every corner of the cave, carefully searching every crevice to eliminate any hidden residual drug-resistant antigens, thereby preventing any future hidden dangers of relapse. Inside the villagers at the foot of the mountain, the drug-resistant antigens were gradually cleared away by the brand-new immune response. Their high fevers broke, their coughs stopped, and their organ damage slowly began to repair. As the battle subsided, Tang Sanzang gathered everyone and the villagers, calmly explaining the knowledge surrounding drug resistance. "Drug-resistant antigens originate from genetic mutations in pathogens, resulting in immune escape. The old immune response fails to recognize them, and their pathogenicity grows stronger. Do not abuse anti-infective medications; protecting your own immunity is the best way to reduce the occurrence of drug resistance. Regular immune monitoring is necessary to detect drug-resistant antigens early and prevent infections from spreading on a large scale." Sun Wukong scratched his head, deeply moved. "After this battle, I finally understand. When exterminating demons, you must pull them out by the roots. You absolutely cannot leave even a small remnant behind, otherwise they will hide, mutate, and cause a massive disaster all over again." The group rested briefly that night. Early the next morning, they bid farewell to the villagers of Hidden Fog Mountain and continued their journey eastward. Ahead lay Flower and Fruit Mountain, where a trial representing the central immune organs was waiting for them. ________________________________________ Public Education: Immunity Explanation Notes •	Drug Resistance and Immune Escape: Under the pressure of medications (like antibiotics or antivirals) or the body's immune system, pathogens can undergo genetic mutations. This changes their antigen epitopes (their structural "appearance"). As a result, they can evade recognition by the existing immune system—a phenomenon known as immune escape, leading to drug resistance. •	Failure of Old Memory Cells: Because the pathogen's structure has changed, the old Memory T cells and Memory B cells from previous infections cannot recognize the new mutated sites. •	Initiating a New Adaptive Response: To fight the mutated pathogen, the body must launch a brand-new adaptive immune response. The T-cell receptors (TCR) must undergo adaptive changes to recognize the new threat, and B cells must generate entirely new antibodies targeted specifically at the newly mutated epitopes. •	Antibiotic Misuse: The abuse or improper use of antimicrobial drugs acts as a strong selection pressure, rapidly accelerating the mutation and creation of drug-resistant pathogens (superbugs). It is crucial to use these medications only as prescribed.

With a mighty roar, Wukong led the three princes and charged fiercely toward the cave.

My brand-new set of killing skills was made specifically to deal with drug-resistant demons like you!"

The brand-new specific antibodies produced by Sha Wujing arrived at lightning speed, directly neutralizing the immunosuppressive toxins released by the Leopard Demon, rendering them instantly ineffective.

We absolutely cannot leave any remnants behind again!"

Wukong leaped high into the air, focusing all his power into the Golden Cudgel, and delivered a devastating blow squarely onto the critical mutated antigen epitope of the Leopard Demon.

Regular immune monitoring is necessary to detect drug-resistant antigens early and prevent infections from spreading on a large scale."

Sun Wukong scratched his head, deeply moved. "After this battle, I finally understand. When exterminating demons, you must pull them out by the roots. You absolutely cannot leave even a small remnant behind, otherwise they will hide, mutate, and cause a massive disaster all over again."

The group rested briefly that night. Early the next morning, they bid farewell to the villagers of Hidden Fog Mountain and continued their journey eastward. Ahead lay Flower and Fruit Mountain, where a trial representing the central immune organs was waiting for them.

Public Education: Immunity Explanation Notes

• Drug Resistance and Immune Escape: Under the pressure of medications (like antibiotics or antivirals) or the body's immune system, pathogens can undergo genetic mutations.
